Ipriflavone is a drug that is used mostly for the treatment of bone diseases in women. The Ipriflavone drug is synthesized in a laboratory. It’s made from a substance called daidzein. Daidzein is obtained from the soy plant. Ipriflavone treats and prevents osteoporosis (weak bones), especially in old age women. It treats bone disease Paget's and prevents osteoporosis caused by certain medications and relieved the pain associated with osteoporosis. This drug is also used to increase metabolism. Bodybuilders usually use this drug to increase their metabolism.

What is Ipriflavone?

The formula of Ipriflavone is 7-isopropoxy-3-phenyl-4H-1-benzopyran-4-one, 147. Ipriflavone is derived from naturally occurring isoflavones. The chemical formula of it is C18H16O3. It is used to prevent osteoporosis, help in the maintenance of bone density and degrade bone resorption. This drug is prescribed too early postmenopausal women aged between 40 to 48 years.

After 12 months of this medicine intake, there will be a 1.1% improvement in bone density of the lumbar spine. After twenty-four months of treatment, bone density will improve up to 1.2 %.

Ipriflavone is devoid of estrogenicity as it does not induce secretion of LH in postmenopausal women. It can effectively improve bone density when combining with low doses of estrogens. It also interferes with osteoclast formation.

Ipriflavone allows the osteoblasts to formed the bone mass by slowing down the action of osteoclasts. Osteoclasts are bone-eroding cells. So when actions of these cells are slowed down, the bone-forming cells (osteoblasts) will easily build up bone mass.

As a result of much research, it has been found that both ipriflavone and estrogen have a similar chemical structure. Due to similar structures, ipriflavone mimics the action of estrogen on the prevention of loss of bones. Estrogens hormones inhibit the activity of osteoclasts that causes bone resorption.

Ipriflavone does not show any same effects on the pituitary gland or hypothalamus as estrogens.   • Alendronate can reduce osteoplastic bone absorbing activity. Alendronate is a bisphosphonate. It is effective for women who cannot bear estrogen therapy. It helps in the prevention of bone loss in women after menopause.
  • Another drug that helps in decreasing bone resorption is Calcitonin. It is an expensive drug. It has to be taken in the form of injection.
  1. The natural alternative, Ipriflavone

Ipriflavone is a 7-isopropoxy iso-flavone. It is synthesis by isoflavone. It is best for the treatment to suppress the resorption of bone. So, ipriflavone is the best drug to prevent bone resorption than the drugs mentioned above, as it has no side effects. Experiments and researches show that ipriflavone show higher activity of bone-building in the body when its intake with other supportive drugs. Ipriflavone is a registered drug for the treatment of osteoporosis in Japan and many countries of Europe. Ipriflavone is an alternative for hormone and other drugs in the treatment of osteoporosis and increases bone mass.

The intake of ipriflavone with some dose of estrogen can increase anti-osteoporotic effects. Thus proved that ipriflavone is better than other synthetic drugs. It is cheap, easily available, and has fewer side effects than other drugs such as Calcitonin, sodium fluoride, Raloxifene, and Alendronate. Ipriflavone is a naturally occurring drug, and it cost very less amount to make ipriflavone from iso-flavone.

  1. Ipriflavone and vitamin D

Vitamin D performs the function of absorption of calcium from the gut to the bones' resorption. The calcium is an essential nutrient of bones. Calcium makes the bone strong. The deficiency of calcium can cause many bone diseases, including osteoporosis—Vitamin D plays a major role in the absorption of calcium in bones. So deficiency of vitamin D can lead to the development of osteoporosis in the body. The liver metabolizes vitamin D in the body. The failure of metabolizes of vitamin D in the liver is also the main cause of deficiency of calcium in bones.

The intake of both vitamin D and ipriflavone can act more effectively and efficiently for increasing bone density. Results of experiments show that bone-building activity accelerates when ipriflavone and vitamin D3 are taken together compared to the single injection of ipriflavone. The injection of vitamin D3 alone also does not affect. The combination of both increased the mineral density of the bone in the full length of the femur.

It was found out that vitamin D3 and ipriflavone proves effective is halting of loss of bones in postmenopausal women. The average loss of bone by injection of the combination of vitamin D3 and ipriflavone was only 0.33 %. In contrast, the loss of bone was 2.37 % when ipriflavone was injected alone, and bone loss for vitamin D3 alone group was 1.15 %.

A study of 2 years revealed the effects of ipriflavone bone-building spine in postmenopausal women who have a low density in vertebral bone. The women took 200 mg of ipriflavone with one gram of calcium three times a day.

After six months of these intake of ipriflavone and calcium combination, the density of these women's spinal bone increases by 1.4 %. The women placed in placebo who only consumed calcium and placebo, their density of bone decreased by 1.2 % in 2 years. The women who recently become menopausal decreased their bone density by 4.9 % during the second year. The study's result was the most rapid loss of bone density experienced during the first five years of menopause.

Ipriflavone can stop bone loss, which occurs shortly after menopause. An experiment examined this ability of ipriflavone. In the experiment, about 50 postmenopausal women with low density in vertebra bone inject 200 mg of ipriflavone three times daily along with calcium. The other postmenopausal women received a placebo. After two years, the result shows that women who intake both ipriflavone and calcium experience no loss in vertebral bone density.

  1. Ipriflavone have stronger actions than calcitonin

Many researchers compared the actions of ipriflavone with FDA approved osteoporosis treatment hormone called Calcitonin. Researchers conducted many experiments to find out which one is ideal for the treatment of osteoporosis. Those experiments show that ipriflavone has a greater ability to resorption bones and inhibit the formation of bones.

Ipriflavone also has greater power to stimulate the osteoblasts to form the new bone cells. These results prove that ipriflavone in stronger in actions then calcitonin hormone. Ipriflavone has no serious side effects, while Calcitonin found to have more serious side effects on the human body.

  1. Ipriflavone and vitamin K

The human body requires vitamin K to convert inactive proteins of bone called osteocalcin into an active form that supports calcium into bones. Vitamin K plays many essential roles, such as halting osteoclasts' actions, which leads to preventing the formation of new osteoclasts. Vitamin K also plays an important role in the metabolism of bones.

The combination of ipriflavone and vitamin K leads to better functioning and mobility of bones. Experiments on actions of vitamin K on bones show that it has the same inhibitory effect on resorption of bones as ipriflavone. Still, the mechanism of both ipriflavone and vitamin D is different. The combination of ipriflavone and vitamin K acts effectively on the prevention of resorption of bones.

Precautions and side effects of ipriflavone

Ipriflavone is a safe drug if it intakes with proper supervision and on the advice of a doctor. It does not have any serious side effects. It is suitable for most people. Some of the side effects of ipriflavone are:

  • Dizziness
  • Stomach pain
  • Diarrhea

Some research demonstrated that ipriflavone could cause decreased white blood cells if the patient takes it for longer, like six months or more. The decreased in the number of white blood cells is called lymphocytopenia. So it is required that the number of white blood cells is monitored in patients taking ipriflavone for a longer time.

Following precautions should be taken

  • Weak immune system

 It is recommended that people having a weak immune system should not intake ipriflavone without consulting with a physician. The intake of ipriflavone decreases white blood cells, which affects the immunity of the body. The immunity of the body decreases by a higher intake of ipriflavone. If one's immune system is already weak, he/ she will intake ipriflavone for a longer period; then, his/ her body will lose the ability to fight off infections.

  • Lymphocytopenia

As mentioned above, ipriflavone affects the number of white blood cells in the body, so it is concerned that longer and higher uptake of ipriflavone can cause lymphocytopenia or it might affect the pre-existing lymphocytopenia.

  • Breastfeeding and pregnancy

 It is not safe to use ipriflavone during pregnancy or post-pregnancy. It can affect the breastfeeding of a mother. So it is recommending that it should be avoided during pregnancy.

Dosing of ipriflavone

It is recommended that the dosing of ipriflavone should be followed, as suggested by your doctors. As a result of a scientific study following doses are studied for uptake:

  • After postmenopausal treatment of weak bones, 200 mg of ipriflavone three times a day will be best.
  • 600-1200 mg daily uptake of ipriflavone will be okay for the treatment of Paget's disease.
  • For the treatment of renal osteodystrophy, 400 to 600 mg of ipriflavone daily should uptake.

Tips for up taking the ipriflavone

The procedure that should be followed for the intake of ipriflavone is the same as other medicine. However, the following things should be followed while in taking pills of ipriflavone:

  • Never take ipriflavone without consulting from a doctor.
  • Always take medicine prescribed by the doctor.
  • Take the right amount of medicine as prescribed by your physicians at the right time.
  • If the medicine is not showing the effective result or are still feeling the pain of osteoporosis, consult it again.
  • Never take ipriflavone during pregnancy. However, if your doctor suggested them, then take it in less amount.
  • Keep the medicine at room temperature and far from the approach of children.

It has been concluded that ipriflavone is a drug that is synthesized from iso-flavone hormone found in soy plants. Ipriflavone has the same chemical structure as estrogen. Ipriflavone drug increases the calcium absorption in bone and decreases the breakdown of bones. Ipriflavone is used mostly for the treatment of osteoporosis in postmenopausal women.

 It relieves the pain caused by osteoporosis and makes the bones stronger. Ipriflavone combination with different bone supporting elements such as ipriflavone combination with calcium, ipriflavone with vitamin K, and it’s with vitamin D3 increases its activity, becoming more effective when patient inject it with these bone supporting elements. The combination of ipriflavone with bone supporting element is called the ipriflavone complex.

It prevents loss of strength of bones, stimulating action of osteoblasts and improves the effect of estrogen to prevent osteoporosis. When ipriflavone is used with estrogen, it allows fewer estrogen doses to be used in postmenopausal women. The main uses of ipriflavone drugs are in preventing and treating weak bones in postmenopausal women. It helps in the reduction of pain associated with osteoporosis.
